On 17 November 1836, she married James Miller Tulloch in Blount County, Tennessee. He was the son of John L. Tulloch.
Mary Best and James Tulloch had nine children, six boys and three girls. After about twenty-two years of marriage, James Miller Tulloch died on 12 March 1859 in Blount County. He was tried at Baker's Creek Cemetery. Mary outlived her husband by almost forty years. She died on 10 May 1899. She was eighty years old and had lived all her life in Blount county. She is buried at Williamson's Chapel Cemetery in Blount County, Tennessee with the rest of her Best kinsmen.
