Ann was the eighth of the twelve children of Mary (Smith) and Lawrence Roelofson, Jr.

On October 22, 1830 in Tremont, Tazewell, Illinois, she married John Tucker Scott and they had 12 children:

The Tucker Scott family left Tazewell County, Illinois for the Oregon Trail on April 2, 1852 with a total of 27 people and five green and yellow covered wagons. This included eleven members of the Scott family and Ann's sister Martha with her husband Levi Caffee and their two boys. The group also included a cousin, William Goudy, with his sick wife and baby, John Goudy and 5 young men as drivers. One of the three additional travelers was a photographer.

Tucker Scott's Bible has an unidentified newspaper clipping glued in it that says, in part: "Departed this life, on the plains, about 30 miles west of Ft. Laramie, on the 21st of June, 1852,[note: she died Sunday June 20 and was buried on the 21st] Mrs. Ann Scott, consort of John Tucker Scott, late of Tazewell County, Ill... She was attacked by cholera, which finished its awful work in nine short hours. She expressed in her last illness a calm confidence in the Redeemer, and a resignation to his will. She left a husband and nine children to mourn her loss, which in this world can never be repaired."
