Rowland, sometimes called, Roland, was born in Middlesex, England in 1812. In 1821, Rowland Brotherhood began working for his father on road building and other ground engineering contracts. He married Priscilla Penton (daughter of William and Mary Ann Penton) in 1835 and they had 14 children.

Along with his father, he obtained a contract for work on part of the construction of the Great Western Railway, building the foundations of Wharncliffe Viaduct at Hanwell. He worked for Mr Brunel on the Great Western Railway. While Brotherhood is best remembered for his work for the Great Western Railway, he also ran a large engineering works at Chippenham, producing railway components and tools, wagons and carriages, and iron bridges.

Their eldest son, Rowland Jr. (b. 18 November 1841) played Cricket for Gloucestershire. He married Julia Fanny Cambridge and they had 8 children.

Most of their sons were also engineers like their father. Rowland Brotherhood Sr.'s second son was Peter (1838-1902). They also had a son named Stanley (1880-1938).

Rowland Brotherhood Sr. died from an illness at his home in Bristol on 4 March 1883, and is buried there in Arnos Vale Cemetery.
