John was born between 1828 and 1832, probably in North Carolina, a son of Dr. Jesse Isler (~1796–1865) and Mary A. E. Bryan (~1807–1860). In 1850 he was attending the Western Military Academy in Nicholas County, Kentucky. On 21 Dec 1852 he married Miss Bob (Robert Rivers) Meriwether, daughter of Richard T. Meriwether and Elizabeth Rivers, in Obion County, Tennessee. They were the parents of one son, Richard Meriwether Isler (1854–1925). A newspapers article dated 13 Nov 1855 states that John P. Isler fatally shot himself, accidentally, "Tuesday last" while hunting near Memphis. He was buried in the Cronanville Cemetery.

Painful Accident.—John P. Isler accidentally shot and killed himself near Memphis on Tuesday morning last, while hunting. Mr. Isler was at one time a resident of Memphis, but more recently has been living on the river about 100 miles above.
