Nathaniel was born and reared at "Longwood", Farmville, Prince Edward County, VA. He matriculated as a medical student in the 1848/49 class at the University of Pennsylvania with Dr. William F. Carrington, his brother-in-law, as his tutor. Nathaniel also studied medicine in Paris and London hospitals and became a surgeon in the CSA Army.

He married Tennessee Marr in Paris, France and they had two children: Mrs. Mary Scott V. Taliaferro and Mrs. Tennessee Marr V. Edmundson.
