Charles Cox was the son of Thomas Cox (b. abt 1736 in PA/d. 1809 in OH) and his wife, Sarah Davis (b. abt 1737 in PA/d. 1813 in OH). Both of his parents moved with their respective families to central North Carolina in the mid-1700s and were among the first Quakers to settle the area which was then known as Orange County, in the Province of (North) Carolina, British Colony in America. Although these families' home sites did not change, the designation of that area did as in later years the larger counties of Orange and Rowan were divided into new counties; first in 1771 when Guilford County, NC was formed from that part of the original Orange County, and again in 1779 when Randolph County, NC was formed from that part of Guilford County. Therefore, Charles Cox was born in what today is known as Randolph County, NC but, at the time of his birth, was known as Orange County, North Carolina.

On the 21st of July, 1785, in Randolph County, North Carolina, Charles took Amy Barker to be his wife in a Quaker marriage union held at Holly Spring meeting house. It was in Randolph County that their children were born - four daughters (Hannah, Sarah, Amy, and Elizabeth) and six sons (Jesse, Thomas, Enoch, Charles, Isaac, and Nathan).
