Isaac is the son of George Glover and Rebecca Bynum. Census records note Isaac's birthplace as Tennessee; It is interesting that notes written by grandson J.L. Glover note Isaac's birthplace as Tuscumbia, AL, which is on the Tennessee River, a town that incorporated in 1820 shortly after Isaac's birth and is the county Seat for Colbert County, Alabama.

Isaac and his siblings were raised in Blount County, Alabama where their father was a farmer. Their lives are documented in the 1916 book "Historical Sketches of the Bynum Family" by J.W. Bynum, Oneonta, Alabama. Isaac married Dicie Ellis in Benton County, Alabama, on January 15, 1839. Isaac was a farmer. Isaac and Dicie had 9 children. In the 1870's, following the Civil War, Isaac and Dicie's children began to move west to Texas. This was a very common trend at that time and many Blount County locals were moving west.

Son Alfred had died in Texas by accidental gunshot wound in 1871. In 1880, their children Louisa, Phoebe, Green, Eli and Uriah Glover can be found in Ellis County; several moved to Callahan county, Texas, in about 1900. It appears that Isaac and Dicie followed their children to Texas. Isaac died in Texas in 1887 and Dicie died in Callahan County, Texas, in about 1917. Dicie is buried at the Eula Cemetery in Callahan County. They had over 65 grandchildren. Their descendants are numerous.

Isaac Glover's grandson Elzel Lorenzo Glover died in Gainesville, Texas and is buried in Rosston Cemetery according to a third great-grandson Douglas Micah Vail.
