Isaac Sherman Painter, younger son of Barbara Ann (née Hillsberry) and Isaac Painter, senior, was born in 1867 on his family's farm near Houstonville, a now-extinct village in East Bend Township, Champaign County, Illinois. He married Emma Bier on 31 August 1893 in Urbana, Champaign County. The couple became the parents of six children: the first five born in Illinois; the last in Michigan.

Isaac Sherman Painter farmed in the county's East Bend and Condit Townships until 1913, when he moved his family—including his elderly father—to Gratiot County, Michigan. There, Isaac Sherman Painter entered into an farming agreement with Thomas Ogden. Painter would supply labor and capital for two-thirds of the profit; Ogden would supply the land for one-third of the profit.*
