Samuel Mitchell is a son of Judge David Mitchell (b 1761) and Martha 'Mattie' Black (b 1764) from Pennsylvania. The extended Mitchell family moved from Lancaster County, PA to the Ohio wilderness in 1799 when Samuel was about 14 years old.

About 1806 in Ohio, Samuel Mitchell married Hope Bishop from Culpepper County, VA. Before 1830, they migrated to Clinton County, Indiana with their daughters: Martha Black Mitchell (Seawright) and Harriett C Mitchell (Wallace).

Samuel was one of the first Associate Judges in Clinton County. He and his son-in-law, Wilson Seawright engaged in the mercantile business under the firm name of Mitchell & Seawright.
