Samuel Carter II was the seventh child of Samuel and Mary (Barnes) Carter. He was reared at the Carter Mill and was trained as a farmer as well as a miller. When his father died in 1804, Samuel and his brother, Mordicai, inherited the land and the mill.

On 11 Nov 1795, Samuel married Rachel Kemp, daughter of Joseph and Welmett (Haworth) Kemp. They had two children, John and Susanna.

Rachel died between 1800 and 5 June 1805 when Samuel married Edith Newlin. Samuel and Edith had fourteen children: Mary, Sarah, Eunice, Asenath, Ruth, Eli, Era, Samuel, Modicai, Naomi, Edith, Ella, Joel D., and Newlin B.

In 1823 Samuel sold land and interest in the mill in North Carolina and moved to Indiana. Samuel and family settled about two and a half miles south of Plainfield.
