Samuel was the son of Reverend Henry Smith, and his second wife, Dorothy (Cotton), he married Mary Ensign, daughter of James and Sarah (Elson) Ensign, about 1662.

He had a four-acre home lot adjoining Henry Smith in Wethersfield on the south, which he sold to Samuel Boardman. He removed to Northampton, Mass about 1666 to Hadley, Massachusetts about 1680. It is unknown where he was buried but it was likely in Old Hadley Cemetery, which was established in 1666, where is son, Preserved Smith was buried.
