He was born at Jordans, nr Ilminster, Somerset, on the 7th of April 1798 and was the son of William Speke (1771-1839) and Mary Speke nee Dickinson (1778-1805) who had married in 1794.
He was baptised at the Church Of The Blessed Virgin Mary, Ashill, Somerset on the 14th of April 1798.
His siblings were:
1. Juliana Barnes nee Speke (1796-1870)
2. George Speke (b.1799)
3. Benjamin Dickenson Speke (1800-1823)
4. Mary Speke (1801-1827)
5. Ann Speke (1802-1805)
6. Revd. Hugh Speke (1803-1856)
7. Sarah Fewtrell nee Speke (1805-1855)
He also had three half-siblings from his father's 2nd marriage to Frances Andrew (1779-1816):

On the 17th of November 1824, at Ilminster, he married Georgina Elizabeth Hanning (1804-1879). She was the daughter of William Hanning (1770-1834) and Harriet Hanning nee Lee (1771-1808) of Dillington Park, Somerset.
They had eight children:
1. William Arthur Speke (1825-1908)
2. Captain John Hanning Speke (1827-1864)
3. Edward Speke (1829-1857)
4. Revd. Benjamin Speke (1830-1881)
5. Georgina Harriet Speke (1835-1913)
6. Elizabeth Speke (b.1839)
7. Sophia Speke (1841-1931)
8. Matilda Speke (1845-1928)
The family resided at 'Jordans' nr Ilminster, Somerset.

He served in the Army as a Captain of the 14th (The Duchess of York's Own) Regiment of (Light) Dragoons, later becoming a wealthy Land Proprietor and inheriting the Jordans Estate following his father's death in 1839. He also served as a local Magistrate.

He died at Orleigh Court, Buckland Brewer, near Bideford, Devon, on the 19th of November 1887, and was buried in the family vault at St Andrew's Church, Dowlish Wake, Somerset.
