Luke Simpson was born 28 November 1781, in North Carolina, probably in Chowan County, to Samuel Simpson and Mary Swain Simpson. The date is found in his father Samuel Simpson's Kirk of Scotland Catechism, where Samuel listed his children and their birthdates. Luke married Margaret "Peggy" Cook on 29 May 1809 in Orange County, North Carolina.

Luke and Margaret had 5 children: Margaret, Mary, Morgan, Lemuel, and Luke. Margaret and Mary did not marry, and sons Morgan, Lemuel, and Luke Simpson died during the Civil War. All three enlisted in in Company K of the 47th North Carolina Infantry Regiment, in the Confederate Service, in March and April 1862. None of them returned from the war. Morgan is known to have been wounded in the wilderness, later dying, in May 1864. It's not know when and where Luke and Lemuel died.
