Isaac was first married to Ruby Pearl Whitley, b. 1889, d. 12 Mar 1922. Mrs. Etheridge was killed in an Atlanta, Birmingham and Atlantic railroad accident. She was traveling along with her brother-in-law Thomas Watson Etheridge and his wife and infant child en-route to a funeral in Woodbury, Georgia when the car in which they were passengers plunged off of a trestle. Ruby Pearl was killed instantly and the others injured.

Isaac Holder Etheridge and Ruby Pearl Whitley were the parents of a daughte, Ruby Jeanette Etheridge.

Isaac was married on 15 Jul 1925 to Mrs. Beatrice Irons Fuller, daughter of Charles Irons. Beatrice was the mother of two sons, James E. Fuller and Charles F. Fuller.
