Jacob was the 2nd oldest of twelve children of Charles and Mary Magdalena (Scholl) Cantonwine. Those children were Michael, Jacob, Margaret, Charles, Catherine, David "Doc", George, Mary, John, Abraham, Samuel, and Syphronia. Only the last child has not been documented.

Charles, the patriarch of the Cantonwine family, immigrated to America from Prussia in 1806, arriving in Philadelphia, Penn., aboard the ship Orlando. The ship's passenger list shows "Chr. Kuntentwine, single man." The name Kuntentwine later evolved to Cantonwine, and Charles appears to be the only surnamed Kuntentwine/Cantonwine to immigrate to America. Eight or so years later, in about 1814, Charles married Mary Magdalena Scholl, daughter of Charles Jacob and Margaret (Fuhr) Scholl, in Bedford county, Pennsylvania.

Jacob married Christena Dilling, daughter of John and Elizabeth Sarah (Acker) Dilling. Jacob and Christena were the parents of thirteen children: Henry Dillon, George Dillon [who both died in the Civil War], Mary Ann "Jane" (m. John Coon), Susan (m. Samuel Stewart), Sarah Elizabeth (m. Joseph Milton Lamb), William Wesley (single), Catherine (m. Casper Stipp), Martha (died in infancy), Alfred Dennis (m. Etta Hannah Guire), Nancy Ellen (m. Thaddeus Duckett), John David (m. Addie Guire), Emma Jane (m. John Burgman), and A. J. G. (daughter, last found, at age 10, living with her brother Dennis Alfred's family in 1880. She most likely died between 1880 and 1885 since she isn't listed with her brother's family in the 1885 census.

"Early settlers in the Nathrop area were Jacob Cantonwine, the Nashes and Royals who came together to Nathrop in a wagon train, which was the first wagon train down Trout Creek Pass, in August of 1866."
