Moses Chapline was the son of William Chapline III (1686-1752) and Elizabeth Riley (1685-1745). He was a resident of Frederick County, Md. (now Washington County). He served in the militia during the French and Indian War.

Moses' older brother Joseph was the founder of Sharpsburg, Maryland. A younger brother, William, lived in the area now known as Shepherdstown, W.V.

In 1740 Moses married Janette Caton.
