William was the son of John Prescott Brooke and Hester Bennett Brooke. At age seventeen he operated the Brooke Ferry across the Etowah River in Chreokee County, Georgia.

He married Mary (Polly) Timmons April 2, 1835. They lived in Cherokee County for several years then purchased land and moved to Bartow County in January, 1844.
