John Chambers was the youngest son of Captain Robert Chambers and his wife, Mary Caldwell Chambers. John was most likely born after his family moved from Middle Spring to the Buffalo Valley, near Sinking Spring about three miles southwest of Mifflinburg.

After his father's death, his mother and older brother, Robert, were required to travel to Carlisle to provide more information on the father's estate.

While they were gone, young John was playing too close to the fire. His clothes suddenly caught on fire and John was badly burned. He did not survive his injuries.

His mother, Mary, wasn't told of his death until immediately before she came back home; her son, Robert, even though he had been told, thought to spare his mother this blow for as long as possible.
