Jacques and his brother Marcq were frères enffans of Jean Le Mahieu (d. 1554), merchant of Lille, and his wife Marguerite Deliot Le Mahieu, daughter of Guillaume Deliot and his wife Catherine Hovine Deliot. Their sister Isabeau Le Mahieu married Robert de Fourmestraux of Lille, her cousin of the third degree, after obtaining a papal dispensation in 1559. Their uncle Nicolas Le Mahieu married Isabelle de Fourmestraux at Lille, October 2, 1548. The Mahieu, Deliot, and Fourmestraux families were part of a group of leading Lillois merchants who were being increasingly disenfranchised and excluded from the magistracy because of their suspected Calvinist sympathies and who were being spied upon by a secret agent of the regent Margaret of Parma.

The arms of this family were d'azur au chevron d'or, accompagné de trois étoiles à cinq rais d'argent .French Walloon. Married 2nd in 1603 to Jane/Jennie "of Canterbury" and all children born in Canterbury. The family fled England to Leyden, Holland to escape persecution. After Jacques death his widow and several children removed from Holland to America.
