Samuel Robert John was born on January 6, 1841, at Brookville in Franklin County, Indiana, to Robert John (1841-1861) and Martha (Wiles) John (1806-1891). Of their three children, Samuel Robert John was the youngest. His two older sisters are Ellen Goodwin (John_ Adair (1833–1905) and Lydia Ann John (1835–1840).
During the American Civil War, CPL. Samuel Robert John served as a volunteer Private in Company "F" of the 3rd Regiment of Ohio Infantry in the U. S. Army. Fifteen days elapsed between Samuel's death at the age of twenty on July 6, 1861, and his interment on July 21, 1861, in Hamilton, Ohio, at Greenwood Cemetery in Section 1st-East, Lot 546. CPL. Samuel Robert John is reported to have been discharged from service on July 5, 1861, perhaps because of severe wounds or illness. His death came one day later.

His name is among the several thousand names inscribed within Hamilton's Soldiers, Sailors & Pioneers Memorial erected in 1904 primarily to honor U.S. military veterans. The Memorial, depicted here in Bruce Garver's photo of Nov. 4, 2017, is Hamilton's largest and most impressive structure designed primarily to commemorate the several thousand Hamiltonians who served their country to suppress rebellion and to eradicate slavery during the American Civil War.
