Jacob Rothrock Kistler was the fifth of nine known children of Jacob Kistler (1775-1844) and Catherine Rothrock (1777-1843), who married c1796 in Pennsylvania.

Jacob first married to Mary E. (Evaline or Elizabeth?) Gruber (1812-1866) on 21 Mar 1833 at Marion Township, Marion County, Ohio. Jacob and Mary were the parents of eleven known children, namely:

1. Christian George Kistler (1834-1895)
2. Sarah Ann Kistler (1835-1835)
3. George Rothrock Kistler (1837-a1880)
4. Austin Justis Kistler (1839-1922)
5. Louisa Arabella Kistler Anderson (1841-1897)
6. Catherine Elizabeth Kistler Anderson (1843-1931)
7. Harriet Ellen Kistler Bulleit (1845-1905)
8. Sarah Frances Kistler Harbaugh (1847-1918)
9. Mary Ann Kistler Elbert (1850-1924)
10. Minerva Etta Kistler Beers (1852-1918)
11. Jane Kistler (???-???)

After Mary died, Jacob reportedly remarried to Sarah Ellen Kistler (1842-1885)
