Josiah Cowles was a son of Calvin Cowles and his wife Miriam Atwater, of Wolcott, Connecticut. He was twice married, first to Deborah Sandford, of Merien, Connecticut in 1815, and second to Nancy Caroline Carson, the widow Duvall, in North Carolina in 1828.

There were four children born of the first marriage: Eliza Ann Cowles (30 April 1817), Elvira Deborah Cowles (18 Jan. 1819), Calvin Josiah Cowles (6 Jan. 1821, born in NC), Abel Sanford Cowles (1 Oct. 1824).

There were seven children born of the second marriage: Josiah Cowles, Jr. (17 Dec. 1829), Andrew Carson Cowles (12 Jan. 1833), Miles Melmouth Cowles (16 April 1835, CSA soldier), Lewis William Cowles (3 Dec. 1837), William Henry Harrison Cowles (22 April 1840), Henry Clay Cowles (17 June 1842) and Caroline Mary "Carrie" Cowles (9 Oct. 1845-20 March 1913, m. Captain M. M. Teague, CSA).
