Carlo Emmanuel was the son of King Vittorio Amadeo II of Sardinia and his wife Princess Marie-Anne of France.

On 15 March 1721, he married Princess Anne Christin Luise of Sulzbach. They were the parents of Vittorio Amadeo Teodoro who died as a child. Anne died due to complications of childbirth.

On 20 August 1724, Carlo married as his second wife Princess Polixene Christin of Hesse-Rheinfels. They were the parents of 6 children-King Vittorio Amadeo III, Maria-Leonora, Maria-Luisa Gabriella, Maria-Felicita, Emmanuel Filiberto, Duke of Aosta, and Carlo Francesco, Duke of Chablis. Emmanuel and Carlos died as children.

In 1737, he married as his third wife Princess Marie-Elisabette Terese of Lorraine. They were the parents of Carlo Francesco, Duke of Aosta, Maria-Vittoria, and Benedetto Maria Maurizio, Duke of Chablis.
