Wilhelmina was the daughter of August Willem, Prince of Prussia and Luise Emelie, Princess of Brunswick-Luneberg. She was born in the Monjibeau Palace, Berlin, Brandenberg, Prussia (now Germany).

On 4 October 1767 in Berlin, Wilhelmina was married to Willem V, Prince of Orange-Nassau, the last Dutch stadtholder.

Wilhelmina and Willem were the parents of Fredericke Luise Wilhelmina, Willem Georg Frederick, Willem Frederick (future King Willem I and the founder of the present Dutch monarchy), and two stillborn infants.
