Knight, in right of his wife of Raglan, Monmouthshire, of Talgarth, Breconshire, Daglingworth, Gloucestershire. Governor of Tretower Castle.
Younger son of Maurice de Berkeley and Elizabeth le Despenser. Grandson of Sir Thomas de Berkeley and Margaret de Mortimer, Hugh le Despenser and Eleanor de Clare.
Sir James married Elizabeth Bluet, daughter and heiress to Sir John BLuet and Katherine Wogan or Gwrgan, widow of Bartholomew Picot of Wilkesworth who died in 1388. They had two sons, James 6th Lord Berkeley and Maurice. After his death, Elizabeth remarried a Sir William Thomas.
