Henry Spencer Barns was a son of Edward Howard Barns and Mary Ann Letcher Barns. It is believed he had four full-siblings. He had six half-siblings from his father's second marriage to Ann/Anna Beard Barns. He also had two half-siblings from his mother's second marriage to Simeon Howard Case.

Henry Spencer Barns married Sarah Ellen Nelson on July 4, 1878 in Winfield, Cowley, Kansas per a letter from J. W. Cottingham, Probate Judge, Cowley County, Kansas. They had six children.
