John was a son of Rev. Eli and Anna (Binion) Wilson Barbre, and was the last surviving member of his father's family of nine children.

He married Margaret R. McKee 5 March 1857 in Sangamon County. They were the parents of six children: James A. who married Martha E. Coe; William E., married to Emma J. Williams; Edwin H.; Anna L.; and Jennie Barbre White; and George Barbre.
