She was the seventh child and fourth daughter of King Friedrich Wilhelm I of Prussia (1688-1840) and Sophia Dorothea of Hanover (1687β1757).

On 10 November 1734 in Potsdam, Sophia Dorothea married her Hohenzollern kinsman Friedrich Wilhelm, Margrave of Brandenburg-Schwedt (1700-1771). They had five children.
She died only 46 years old from complications of anasarca.
She and her huaband are buried in identical coffins, Sophie's is on the left, Friedrich's on the right.
