On October 3, 1903, Manning married Nora Orr.
He was best-known for his law enforcement activities, including being appointed as U.S. Marshal in 1937, a position he held until his retirement in 1953. Prior to this position, he had been a deputy sheriff and for a three-year period was the President of the Poughkeepsie Board of Police Commissioners. He was also known for owning and using bloodhounds in criminal and missing person cases.
He also engaged in the real estate business, including constructing many buildings on Academy Street. For a while, he established and operated a taxicab business.
He and his wife had seven children: sons Manning Jr., Raymond and Paul (who died as an infant), and daughters Helen (Mrs. Clayton Todd), Isabel (Mrs. Otto Varrelman), Mary Jane (Mrs. Charles Chandler) and Shirley (Mrs. Theodore Clark).
