His parents were married on September 3, 1660 in London and their first child, Charles, was born October 22, 1660 at Worcester House, The Strand, London. He was baptised on January 1, 1661 at Worcester House, but he died from smallpox on May 5, 1661 at Whitehall Palace. He died before the letters patent for the title, Duke of Cambridge, had been officially conferred. He was buried in Westminster Abbey on May 6, 1661.

The burial of the decedent within Wesminster Abbey on 6 May 1661 is confirmed by the ultimate contemporary source for the burial, that being The Marriage, Baptismal, and Burial Registers of the Collegiate Church Or Abbey of St. Peter, Westminster, Edited and Annotated by Joseph Lemuel Chester (London, 1876), page 154. It states:

"Charles, Duke of Cambridge, son to the Duke of York (future King James II): in a vault on the South side of K. H. 7th's Chapel."
