Daniel Thomas graduated at Yale College in 1733 with the degree A. M. After graduation he went to England and studied "chirurgery" in St Thomas's Hospital in 1737. He returned to Norwich, where he became a prominent businessman. He and his brother Joshua built a wide reputation as importers of drugs, and built large estates for their day.

Daniel Thomas married Jerusha Talcott, daughter of Gov. Joseph Talcott, on December 13, 1744. "In the language of Mrs. Sigourney, she was 'a lady of noble bearing, cultivated intellect, and eminent piety. Though far advanced in years, when I first beheld her, time had not impaired either her physical or mental system. Her tall, majestic form was unbowed, her step elastic, and her heart in ardent, healthful action.... Dr. Lathrop was one of the most liberal of the sons of Norwich in devising for the prosperity of the church and the improvement of the town. His own mansion was a model of elegance beyond anything which had before been built.'"

His will names his wife; son Joshua; Mrs. Abigail Wadsworth; Daniel Huntington of Woodbury, son of his deceased friend; Ezekiel Huntley and Lydia Howard; Daniel Lathrop Coit; Daniel Lathrop, son of his brother Joshua; Yale College; First Society of Norwich; and Norwich School.

Jerusha's will, dated December 9, 1799, names her brother Mathew Talcott; grandchildren of her brother Samuel Talcott; 3 children of her sister Hooker: James, Horace, and Eunice Ellery; William, grandson of her brother Samuel Talcott; Eunice and Elizabeth, daughters of her sister Wadsworth, deceased; Esther Russel, wife of Thaddeus Learned; Daniel L Coit; Thomas Coit, son of Brother Thomas Coit; heirs of Joshua Coit; children of Elizabeth Leffingwell, deceased; Daniel Lathrop and Lydia Austin, children of Brother Joshua; Daniel, son of Brother Joshua; Jerusha, daughter of Thomas; Hannah Huntington, granddaughter of Brother Joseph Coit, deceased; Ezekiel Huntley; First Society.
