Isaac B. Hickerson was born to John Hickerson and Patsey Loving. It is estimated he was born about 1786. His siblings were William Loving Hickerson, John Hickerson and Tabitha Hickerson. In 1793, Isaac and his brothers were given a preemption grant of 640 acres of land on the west branch of Goose Creek in Sumner County, Tennessee, as the young heirs of Captain John Hickerson. Captain Hickerson was killed in early 1791 while serving as a scout with a group of militia in pursuit of a marauding band of Indians on Smith's Fork, a branch of Caney Creek, south of the Cumberland River. Isaac, his mother, and his siblings were all named in his father's probate records.

During the War of 1812, on September 24, 1813, Isaac enlisted as a Private in the Regiment of Cavalry and Mounted Gunmen, Tennessee Volunteers. He was promoted to Corporal, serving in Capt. Evans' Company of Spies with the Mounted Gunmen. He served with his brothers, John and William, in both companies.

Isaac's first wife is unknown but she was enumerated in the 1820, 1830 and 1840 census as a female born about 1794. They had 3 children, Margaret, Moses and Martha. Isaac was enumerated in the federal census in Smith County, Tennessee in 1820, then migrated to Shelby County, Tennessee sometime before 1823, at which time he was summoned for jury duty in Shelby County. In 1826, Isaac took the oath to serve a 2-year term as Constable of Shelby County. He was enumerated in the 1830 and 1840 census in Shelby County. In 1844, he purchased 200 acres of land in Range 8, Section 11 in Shelby County for 12.5 cents per acre, pursuant to a land grant from the state of Tennessee.

It is likely Isaac's first wife died after the 1840 census was enumerated. Isaac then married Mary L. Anderson, who was some 30 years younger than Isaac, born about 1816.

Isaac executed his Will on August 20, 1847. Isaac's Will was probated and recorded in Shelby County on March 20, 1848. In it he named his wife, Mary L. Hickerson, his son, Moses Hickerson, his daughter, Martha (Hickerson) Dalby, and his granddaughter, Martha Smith, who was the daughter of Margaret (Hickerson) Smith, Isaac's daughter who had predeceased him. Isaac's burial location is unknown but is presumed to be in Shelby County, Tennessee.
