Sir John Neville was the son of Ralph Neville, 2nd Earl of Westmorland and Elizabeth Percy, and the grandson of John de Neville and Elizabeth de Holand.

John was the first husband of Lady Anne Holand, daughter of John de Holand, 1st Duke of Exeter and Anne Stafford, before 18 February 1441.

John was invested as a Knight on 01 Dec 1449.

He died before 16 March 1450/51, without issue, as his marriage was supposedly not consummated. His will was dated 1 December 1449 and probated on 5 April 1451, directing his burial to be at Haltemprice, Yorkshire.

His wife, Anne Holand, would marry his uncle, Sir John Clifford, the son of her first husband's grandparents.
