Lady Margaret was the daughter of John Mawle and his wife Margaret of Stoke Nayland. She was born about 1540, probably in that town. On May 26, 1562 in Nayland, Suffolk, she married .John Barker, son of Richard and Alice Barker, who was born about 1540. He was a cloth maker at Nayland, Suffolk. John and Margaret had eleven children baptised at Nayland. He was prosperous, as can be seen by his will, in which he bequeaths five houses to members of his family, and says that other houses have been disposed of. John was buried at Nayland on October 22, 1588, in the churchyard of St. James Church. Margaret also left a will, dated December 24, 1589. She mentions many family members, nephews, nieces, cousins, etc. Margaret was buried on January 11, 1590 in the churchyard of St. James Church at Nayland.
