The eldest of four children born to noted Dr. Frederic E. Humphreys, founder of Humphreys Homeopathic Medicines, and his second wife, the former Frances Maria Sperry, Helen Humphreys, who died at age 45, was their only daughter. She was predeceased by a half-brother, George, in 1868, and a brother, Jay, in 1884. Survivors included her parents and brother Frederick.
