Sir William Spencer Kt, of Wormleighton & Althorp, Northants was the son of Sir John Spencer, Kt, of Snitterfield & Wormleighton and Isabel (Graunt) and was born before 1516. He married Susan, daughter of Sir Richard Knightley, of Fawsley, Northamptonshire. To this union six children were born; Sir John Spencer, Kt, of Wormleighton & Althorp, Isabel, Jane, Dorothy, Anne, and Mary.

Sir William Spencer was invested as a Knight at York Palace (Now Whitehall) in 1529 by Henry VIII. He held the office of High Sheriff of Northamptonshire from 1531 to 1532.
