Shortly after the erection of the first Presbyterian meetinghouse at the Forks of the Brandywine, a piece of land immediately to the east of that building was appropriated for a burial place. During the Old Side–New Side Controversy of the early-mid 18th century, this original meetinghouse and this burial ground were used by the Old Side Presbyterian congregation; those of the New Side congregation formed their own meeting and laid out their own burial ground eastward across the Horseshoe Pike.

The first inscribed headstones placed in this burial ground are those at the graves of Mrs. Jane Sterling and the infant children of Mr. Carmichael, although they were not the first persons buried within this enclosure. These stones were prepared at Burlington, N. J., then the rival of Philadelphia.
Until the mid-to-late-19th century, no record of the interments was kept, and many of those buried there do not have stones to mark their last resting places. It is, therefore, almost impossible to ascertain with precision how many have been buried in this graveyard.

This is the largest of the three graveyards at Brandywine Manor, having been expanded on several occasions.
